First Tissue Cloning Trials

Okay I am planning on trying to clone some plants using this new tissue cloning technique. For those who haven't heard its the process of taking a fragment of a piece of leaf or stem and turning it into an entire plant. I am just now starting to germinate 6 seeds that i will attempt to clone(2xSkunk, 2xBlueVelvet, 2xBubbleGum)
This trial and error begins with new plants. Once they're a couple of weeks old and have about 4 leaves i will snip a solitary leaf off, from each plant, and prepare it to be cloned. Since it takes only a fraction of a leaf ill try and make 3 clones from each leaf. Once they are sterile I will put them into their first jars of agar.
So far i'm not too sure what to use in my agar. I will use the standard agar method(used for mushroom culture) which consist of agar agar, potato flakes, and honey. I'm also going to try and find the growth hormones NAA and BA for this medium. Ba is a hormone that promotes rooting.
A sterile work environment is vital. I am using a homemade glove box for this task.
I won't have any real information ready for this anytime real soon but I will try and keep you informed of everything that happens as it comes.
If anyone has any suggestions or thinks im going about this set up wrongly please let me know.

curiosity mainly? i dont know what that has to do with anything. I mean this is a grow journal right? i am merely documenting my tissue cloning trials so people can read and run trials themselves. Someone asked me to keep a record of these tissue clone trials. If it works i will help explain what you need to know to do it yourself. i have a very limited number of some dank seeds left. i am trying to figure out this whole tissue cloning thing so i can clone my own plants and keep my dank skunk strain alive. Tissue cloning, if done right, is supposed to be convenient. i would like to have the power and option of taking one simple leaf from another growers plant and have the exact same genetics.
If you know more about this then please enlighten me with some ideas.

O well of course! you can make thousands(literally) of clones from a single plant(in its vegetative phase). but not only can u make an infinite number of clones, u can contain them in small jars. this terminates the mother plant strategy. No longer keeping a mother plant in veggie stage all the time. You can easily have 500 copies of the exact same plant in a closet on one shelf. stored and ready to be transplanted when you are.

I was doing a little reading about this.
Have you considered using a part of the plant containing the fastest dividing cells? As in, perhaps a piece of new growth tip on a lower node or something. The highest density of rapidly growing stuff.... dig?
Here's a TC recipe I found that seems pretty generic:
1/2 teaspoon Peter's fertilizer (20-20-20) 2 tablespoon sugar 1 multi-vitamin pill 1 - 2 ml PPM (optional but very hard without it) Agar or gelatin (I've never used gelatin) 4 cups Bottled or RO Sodium Free Water Vinegar or Baking Soda to adjust pH

So, there's a couple more considerations for you... fertilizer would apparently be acceptable in your media, and also the PH should probably be adjusted...

thanks for your research. Ima go ahead and reconstruct my agar media based on the info youve provided. im also assuming i can use distilled water besides RO water. and yeah ill definitely use the newest leaflets from each plant. I was going to wait and do this in a few more weeks but farmer joe had asked me to start this grow journal so i decided to go ahead and get a move on with it. Assuming that this works the first time or two around it will be several months before it is completely finished and full of results. ty for your support

I think a lot of people will be watching this thread.
I wish I had the funds or I would invest in this method, as I'm starting to grow on the idea of Sea Of Green. I like small, fast flowering plants, and in high numbers you can yield very high.
That's awesome that you have the balls to dive right into a new method of cloning. I hear patience is a virtue.

All I can do is try. I honestly don't expect great results the first time around but I will let people know what i am doing either way so they can make their own conclusions based on my findings. So far I've put together a rough media that i will be testing but chances are this won't be the right one. Ive got to go get more supplies sometime after thanksgiving. Also trying to sprout some new seeds but nothing yet. I'll keep you guys posted. Wish me luck!
